Why Culture Cardiomyocytes in a Defined Medium?

Maintaining the health of pluripotent stem cell-derived cardiomyocytes is imperative for generating trustworthy data during downstream experimentation. Culturing cardiomyocytes in serum-free, defined media reduces experimental variability in cell health by eliminating the uncontrolled variables that are often associated with serum-containing media, such as indeterminate levels of vitamins, hormones, and growth factors.

Cardiomyocytes cultured using the Cardiomyocyte Maintenance Media Supplement can also be a part of drug toxicity and small molecule screening workflows where a controlled cell culture environment is essential.

Cardiomyocyte Maintenance Media Use

Human pluripotent stem cells are differentiated into cardiomyocytes using the StemXVivo® Cardiomyocyte Differentiation Kit (R&D Systems, Catalog # SC032B) until widespread beating is observed in the culture. StemXVivo® Cardiomyocyte Maintenance Media can then be used to maintain the differentiated cells for extended culture. Media should be replaced daily.

Note: Repeated mechanical stress can negatively affect cell attachment and survival. For extended culture, daily media exchanges should be handled as gently as possible.

Precautions

This product contains human transferrin. This transferrin was purified from donor plasma and tested at the donor level using an FDA licensed method and found to be non-reactive for anti-HIV-1/2 and Hepatitis B surface antigen.

Detection of Cardiac Troponin T in Human ES-derived Cardiomyocytes.BG01V human embryonic stem cells were differentiated into cardiomyocytes using the StemXVivo®Cardiomyocyte Differentiation Kit (Catalog # SC032B) and maintained in StemXVivo®Cardiomyocyte Maintenance Media Supplement (Catalog # AR011) until day 80 of differentiation. Cardiac Troponin T (red) was detected in cardiomyocytes with a Mouse Anti-Human Troponin T (Cardiac) Monoclonal Antibody (Catalog # MAB1874) and a NorthernLights™ (NL)557-conjugated Donkey Anti-Mouse Affinity Purified Polyclonal Antibody (Catalog # NL007). The nuclei were counterstained with DAPI (blue).

Assay Procedure

Briefly, Cardiomyocyte Maintenance Media is prepared using the following procedure:

  • Dilute 50-fold in RPMI 1640
  • Add 0.5 mM L-glutamine and 1X Penicillin / Streptomycin (optional)
  • Store the media in the dark at 2-8 °C
  • Use within 2 weeks

Reagents provided in the N21-MAX Insulin Free Media Supplement (Catalog # AR010):

  • RPMI 1640
  • L-Glutamine
  • Penicillin/Streptomycin

  • Basal media (e.g., Neurobasal Media from Invitrogen®, Catalog # 21103-029 or equivalent)

Procedure Overview

Dilute 50-fold in RPMI 1640. Add 0.5 mM L-glutamine and 1X Penicillin / Streptomycin (optional). Store the media in the dark at 2 to 8 °C for up to two weeks.
