*The Sample Volume represented is based on the amount of sample incorporated into the reaction after taking into account the assay’s minimum required dilution for a given matrix. Serial dilution may be necessary to achieve some of the final sample volumes represented.

Product Summary

Simple Plex Discovery assay is for the detection of human TNF-Related Activin-induced Cytokine (TRANCE) in cell culture supernatant (CCS), serum, and plasma (EDTA/Heparin) in Sample Diluent SD13.

Precision

Intra-Assay Precision (Precision within an assay) Each control was tested multiple times in one TRANCE assay
Inter-Assay Precision (Precision between assays) Replicates of each TRANCE control were tested in multiple assays performed by at least three technicians using two lots of reagents for human TRANCE

Cell Culture Supernates, Serum, EDTA Plasma, Heparin Plasma

Intra-Assay Precision Inter-Assay Precision
Sample 1 2 1 2
n 16 16 24 24
Mean (pg/mL) 198 9796 253 12325
Standard Deviation 8.65 482 28.1 1286

Recovery

Recovery for the human TRANCE assays was evaluated at three different spiked concentrations of TRANCE protein.

Sample Type Average % Recovery Range %
Cell Culture Media (n=2) 112 107-117
EDTA Plasma (n=2) 98 81-116
Heparin Plasma (n=2) 114 109-120
Serum (n=2) 108 101-115

Linearity

Samples containing and/or spiked with high concentrations of human TRANCE were serially diluted with Sample Diluent to produce samples within the dynamic range of the assay.

Background: TRANCE/TNFSF11/RANK L

TRANCE (TNF-related activation-induced cytokines), also known as RANK Ligand, OPGL, or ODF, is a TNF superfamily ligand produced by osteoblasts, bone marrow stromal cells, and activated T cells. It signals through RANK and is antagonized by the decoy receptor Osteoprotegerin. TRANCE stimulates bone resorption, is required for osteoclast differentiation, and is associated with the development of osteoporosis, vascular calcification, osteopetrosis, and progestin-induced breast cancer. It also mediates body temperature regulation and is essential for the development of a lactating mammary gland.

Long Name:
TNF-related Activation-induced Cytokine
Entrez Gene IDs:

Alternate Names:
CD254 antigen; CD254; ODF; OPGL; OPGLOPTB2; Osteoclast differentiation factor; Osteoprotegerin ligand; RANK L; RANKL; RANKLreceptor activator of nuclear factor kappa B ligand; Receptor activator of nuclear factor kappa-B ligand; sOdf; TNF-related activation-induced cytokine; TNFSF11; TRANCE; TRANCEODFhRANKL2; tumor necrosis factor (ligand) superfamily, member 11; tumor necrosis factor ligand superfamily member 11
