Biological Activity
Key information: NIR Dye s775z, NHS is a near-infrared (NIR) fluorescent dye supplied with an NHS ester reactive group for the labeling of primary amines. Suitable for in vivo imaging.Application: Fluorescence microscopy, in vivo imaging.
Properties and Photophysical Data: When conjugated to antibodies, NIR Dye s775z, NHS is substantially brighter and more photostable than competitor dyes. It retains excellent target specificity, even when the degree of labeling (DOL) is very high. Excitation and emission maxima (λ) are 775 nm and 795 nm, respectively; quantum yield = 0.09; extinction coefficient = 201,000 M-1cm-1; A280 correction factor = 0.11.

Scientific Data
Application of NIR Dye s775z, NHS in HT-1080 human fibrosarcoma. HT-1080 cells were fixed, co-incubated with primary rabbit anti-tubulin overnight at 4 °C. The following day, they were incubated with secondary goat anti-rabbit labelled with NIR Dye s775z (green) for 2 h at room temperature. Cells were co-stained with Hoechst 33342 (blue, Catalog # 5117, Tocris). Dye used at 1:100 dilution. Scale: 30 μm. Application of NIR Dye s775z, NHS in vivo. Mock surgery process of a tumor bearing mouse 3h post retro-orbital co-injection of 100 μL (20 nmol) NIR Dye s775z, NHS conjugated to integrin receptor targeting cRGDfK peptide. Images were taken before the surgery, and after excising the tumor from mouse body.
