Detection of TNF‑ alpha in Human PBMCs by Flow Cytometry. Human peripheral blood mononuclear cells (PBMCs) (A) treated with PMA and Ca2+ ionomycin overnight or (B) resting, were stained with Human Anti-Human TNF- alpha (Research Grade Adalimumab Biosimilar) Biotinylated Monoclonal Antibody (Catalog # FAB9677B) followed by APC-conjugated Streptavidin (Catalog # F0050) and Mouse anti-Human CD3 PE-conjugated Monoclonal Antibody (Catalog # FAB100P). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Adalimumab is a biosimilar directed against the extracellular domain of TNF- alpha. The biotinylated antibody can be used in flow cytometry or various assay format to measure TNF- alpha expression and can be detected by conjugated streptavidin.
