Applications

Recommended Concentration
Sample
Intracellular Staining by Flow Cytometry
0.25 µg/106 cells
See below
Neutralization
Measured by its ability to neutralize TNF‑ alpha -induced cytotoxicity in the L‑929 mouse fibroblast cell line. Matthews, N. and M.L. Neale (1987) in Lymphokines and Interferons, A Practical Approach. Clemens, M.J. et al. (eds): IRL Press. 221. The Neutralization Dose (ND50) is typically 2-12 ng/mL in the presence of 0.25 ng/mL Recombinant Human TNF‑ alpha and actinomycin D.

Please Note: Optimal dilutions should be determined by each laboratory for each application. General Protocols are available in the Technical Information section on our website.

Detection of TNF‑ alpha in Human PBMCs by Flow Cytometry. Human peripheral blood mononuclear cells (PBMCs) (A) treated with PMA and Ca2+ ionomycin overnight, or (B) resting, were stained with Human Anti-Human TNF- alpha (Research Grade Adalimumab Biosimilar) Monoclonal Antibody (Catalog # MAB9677) followed by Goat anti-Human APC-conjugated Secondary Antibody (Catalog # F0135) and Mouse anti-Human CD3 PE-conjugated Monoclonal Antibody (Catalog # FAB100P). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Cytotoxicity Induced by TNF-alpha and Neutralization by Human TNF-alpha Antibody. Recombinant Human TNF-a (Catalog # 210-TA) induces cytotoxicity in the the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by Resazurin (Catalog # AR002). Cytotoxicity elicited by Recombinant Human TNF-a (0.25 ng/mL) is neutralized (green line) by increasing concentrations of Human Anti-Human TNF-a (Research Grade Adalimumab Biosimilar) Monoclonal Antibody (Catalog # MAB9677). The ND50 is typically 2-12 ng/mL in the presence of the metabolic inhibitor actinomycin D.

Background: TNF-alpha

Adalimumab is a biosimilar directed against the extracellular domain of TNF- alpha. This constitutes the immunogen used for the anti-Idiotype catalog numbers MAB9616 and MAB9546. The biotinylated antibody (catalog number IC9677B) can be used in flow cytometry or various assay format to measure EGFR expression and can be detected by conjugated streptavidin.

FAQs

  1. What is a Biosimilar antibody?

    • A Biosimilar antibody is a non-therpeutic, recombinantly-expressed antibody which has the same sequence as the corresponding therapeutic antibody.  It is for research use only. The nomenclature of the Biosimilar antibody includes the name of the therapeutic antibody.  For example, Catalog # MAB9577 is called Human EGFR (Research Grade CetuxiMab Biosimilar) antibody.

  2. Does this Biosimilar antibody have the same sequence as its corresponding therapeutic antibody?

    • This Biosimilar antibody has the same variable region sequence as the therapeutic antibody, but the constant region is not the same. It contains a proprietary human IgG constant region.
