Scientific Data
Detection of Human GSK 3 alpha and GSK‑3 beta by Western Blot. Western blot shows lysates of U2OS human osteosarcoma cell line and HeLa human cervical epithelial carcinoma cell line. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human GSK-3a/ beta Monoclonal Antibody (Catalog # MAB25063) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for GSK-3a at approximately 46 and GSK-3 beta at approximately 51 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
GSK‑3 alpha / beta in HT‑29 Human Cell Line. GSK-3a/ beta was detected in immersion fixed HT-29 human colon adenocarcinoma cell line using Mouse Anti-Human GSK-3a/ beta Monoclonal Antibody (Catalog # MAB25063) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). GSK-3a/ beta was detected in immersion fixed NIH-3T3 mouse embryonic fibroblast cell line using Mouse Anti-Human GSK-3a/ beta Monoclonal Antibody (Catalog # MAB25063) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Preparation and Storage
- 12 months from date of receipt, -20 to -70 °C as supplied.

Background: GSK-3 alpha/beta
Glycogen Synthase Kinase-3 (GSK-3) is a serine/threonine kinase initially identified as an inhibitor of glycogen synthase. Two isoforms (GSK-3 alpha and GSK-3 beta ) share 85% amino acid identity. GSK-3 beta, inhibited by phosphorylation at S9 by Akt, is involved in energy metabolism, body pattern formation, and neuronal cell development.
