Detection of Fibronectin in WS‑1 Human Cell Line by Flow Cytometry. WS-1 human fetal skin fibroblast cell line was stained with Mouse Anti-Human Fibronectin Fluorescein-conjugated Monoclonal Antibody (Catalog # IC1918F, filled histogram) or isotype control antibody (Catalog # IC002F, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: Fibronectin
Human Fibronectin (FN) is a 210-220 kDa extracellular matrix protein that serves as a cornerstone for cell-matrix interactions. It is a sulfated glycoprotein 2055 amino acids (aa) in length. Along its length, it exhibits three types of repeating modules; twelve type I, two type II and 16 type III modules. There are more than 15 alternative splice variants, most in the C-terminal half of the molecule. FN circulates as a disulfide-linked homodimer, a product of hepatocytes. It also exists as a very large covalent multimer, likely the product of a wide variety of cell types. Within these multimers will be found FN-Fibrinogen, -Collagen 13 and -Mac2BP complexes. The multidomain nature of FN allows for binding of Fibrin, GAGs (HS and CS), Syndecans and Integrins alpha 5 beta 1 and alpha V beta 3. Over aa 31-2386 (full-length), human FN shares 91% aa sequence identity with mouse FN.
