Detection of CD8 alpha in Human Blood Lymphocytes by Flow Cytometry. Human peripheral blood lymphocytes were stained with (A) Mouse Anti-Human CD8 alpha Monoclonal Antibody (Catalog # MAB3803) or (B) Mouse IgG2A isotype control antibody (MAB003) followed by anti-Mouse IgG Phycoerythrin-conjugated secondary antibody (F0102B) and Mouse Anti-Human CD3 epsilon Allophycocyanin-conjugated Monoclonal Antibody (FAB100A). Staining was performed using our Staining Membrane-associated Proteins protocol.
Detection of CD8 alpha in HEK293 Human Cell Line Transfected with Cynomolgous Monkey CD8 alpha and eGFP by Flow Cytometry HEK293 human embryonic kidney cell line transfected with (A) cynomolgous monkey CD8 alpha or (B) irrelevant protein, and eGFP was stained with Mouse Anti-Human/Cynomolgous Monkey CD8 alpha Monoclonal Antibody (Catalog # MAB3803) followed by APC-conjugated Anti-Mouse IgG Secondary Antibody (F0101B). Quadrant markers were set based on control antibody staining (MAB003).Staining was done using our Staining Membrane-associated Proteins protocol.
CD8 alpha in Human PBMCs. CD8 alpha was detected in immersion fixed Human peripheral blood mononuclear cells (PBMCs, positive staining) and HDLM‑2 human Hodgkin’s lymphoma cell line (negative control) using Mouse Anti-Human CD8 alpha Monoclonal Antibody (Catalog # MAB3803) at 8 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; NL007) and counterstained with DAPI (blue). Specific staining was localized to cell surface. Staining was performed using our protocol for Fluorescent ICC Staining of Non-adherent Cells.
CD8 alpha in Human Lymph Node. CD8 alpha was detected in immersion fixed paraffin-embedded sections of human lymph node using Mouse Anti-Human CD8 alpha Monoclonal Antibody (Catalog # MAB3803) at 0.5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell surface. Staining was performed using our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.
