Scientific Data
Chemotaxis Induced by CCL14a/HCC‑1 and Neutral-ization by Human CCL14/ HCC‑1/HCC‑3 Antibody. Recombinant Human CCL14a/ HCC-1 (Catalog # 1578-HC) chemoattracts the BaF3 mouse pro-B cell line transfected with human CCR1 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Human CCL14a/HCC-1 (10 ng/mL) is neutralized (green line) by increasing concentrations of Rat Anti-Human CCL14/HCC-1/ HCC-3 Monoclonal Antibody (Catalog # MAB3241). The ND50 is typically 0.5-2.5 µg/mL.

Background: CCL14/HCC-1/HCC-3
CCL14 is a chemokine that promotes chemotaxis of T cells, monocytes, and eosinophils. It occurs in two isoforms resulting from differential mRNA splicing. Following cleavage of a 19 amino acid signal peptide, mature CCL14a (aa 20‑93) is a 74 amino acid peptide that is also known as HCC-1 (Hemofiltrate CC Chemokine-1). It is a weak CCR1 agonist, however, an 8 amino acid N-terminal truncation (aa 28‑93) allows potent signaling through CCR1 and CCR5. CCL14b, also known as HCC-3, is a 90 amino acid peptide (aa 20‑109) resulting from the insertion of 16 amino acids between residues 7 and 8 of CCL14a.
