GMP Human T Cell Media is formulated for the ex vivo culture of human T lymphocytes for preclinical or clinical ex vivo use. This media is optimized for T cell expansion in a G-Rex bioreactor with R&D Systems GMP cytokines, in a variety of combinations (IL-7 and IL-15, or alternatively, IL-2). It is also compatible with different cell activation methods, including particle, bead-based, or plate-bound. It is highly recommended to supplement this product with human AB serum. The activation reagents, culture vessel, and cytokine growth factor combinations used with this media should be optimized by application or experimental protocol.
The bagged format of GMP T cell media (CCM038-GMP-1B) is designed to integrate seamlessly into a closed system workflow. Use with ProPak GMP cytokines and a G-Rex bioreactor for a derisked T cell manufacturing process.

T cell viability and growth are equivalent in fresh and pre-made media stored at 4 °C for at least 5 weeks. Xeno-Free GMP Human T Cell Media (CCM038-GMP-1L) was formulated with 5% human AB Serum (HABS001-GMP-100ML) and 10 ng/mL of IL-7 (BT-007-GMP) and IL-15 (BT-015-GMP) cytokines. Media was either used fresh or stored at 2-8 °C for 7-35 days prior to use in human T cell culture. T cells were expanded with a T cell activator in a G-Rex 6M bioreactor for 7 days. T cell viability (A), cell density (B), and fold expansion (C) after 7 days in culture are shown. Data is the average of 3 purified T cell donors with error bars ± SD.

Purified CD4+ and CD8+ T cells from 3 healthy donors were cultured in a 6M G-Rex Bioreactor in GMP Human T Cell Media (Catalog # CCM038-GMP-1L) with 5 ng/mL of Recombinant Human IL-7 GMP Protein (Catalog # BT-007-GMP), 2.5 ng/mL Recombinant Human IL-15 GMP Protein (Catalog # BT-015-GMP) and either 5% human AB serum or a xeno-free serum replacement. After 9 days in culture, cells were assessed for confluency and viability.

T cell fold expansion and viability with GMP Human T Cell Media are comparable to competitor media. Purified CD4+ and CD8+ T cells from 3 healthy donors were cultured in a 6M G-Rex Bioreactor in GMP Human T Cell Media (Catalog # CCM038-GMP-1L) with 5 ng/mL of Recombinant Human IL-7 GMP Protein (Catalog # BT-007-GMP), 2.5 ng/mL Recombinant Human IL-15 GMP Protein (Catalog # BT-015-GMP) and 5% human AB serum, or a competitor media. After 9 days in culture, cells were assessed for fold expansion and viability.

T cell culture in GMP Human T Cell Media results in lower exhaustion marker expression. Purified CD4+ and CD8+ T cells from 3 healthy donors were cultured in a 6M G-Rex Bioreactor in GMP Human T Cell Media (Catalog # CCM038-GMP-1L) with 5 ng/mL of Recombinant Human IL-7 GMP Protein (Catalog # BT-007-GMP), 2.5 ng/mL Recombinant Human IL-15 GMP Protein (Catalog # BT-015-GMP) and 5% human AB serum, or a competitor media. After 9 days in culture, cells were assessed for expression of exhaustion markers, PD-1 and Tim-3. Data is the average of 3 donors ± SD.

GMP Human T Cell Media supports T cell editing with Lentivirus or the TcBuster Transposon system. Purified CD4+ and CD8+ T cells from 3 healthy donors were cultured in a 6M G-Rex Bioreactor in GMP Human T Cell Media (Catalog # CCM038-GMP-1L) with 5 ng/mL of Recombinant Human IL-7 GMP Protein (Catalog # BT-007-GMP), 2.5 ng/mL Recombinant Human IL-15 GMP Protein (Catalog # BT-015-GMP) and 5% human AB serum. Activated T cells were transposed with the TcBuster transposon system or transduced with Lentivirus to express CD19CAR and eGFP. After 9 total days in culture, cells were assessed for expression of the inserted cargo (eGFP). Data is the average of 3 donors ± SD.
